运动对高脂血症大鼠肝脏胰岛素受体底物-2表达的影响 被引量:5

摘要 目的观察高脂血症大鼠运动后肝脏胰岛素受体底物-2(IRS-2)的变化,探讨IRS-2在运动改善大鼠糖脂代谢中的作用。方法将26只Wistar大鼠分为正常对照组(9只)、高脂膳食组(9只)和高脂膳食+ 60 min运动组(运动组,8只)。测定各组大鼠肝脏IRS-2 mRNA及蛋白表达的变化。结果①高脂膳食组的三酰甘油(TG)、胆固醇(TC)、低密度脂蛋白-胆固醇(LDL-C)显著高于正常对照组和运动组(P值均<0.05),运动组的高密度脂蛋白胆固醇(HDL-C)显著高于高脂膳食组(P<0.05)。②高脂膳食组大鼠肝脏IRS-2 mRNA及蛋白表达水平显著低于正常对照组和运动组(P值分别<0.01、0.05)。结论高脂饮食在导致大鼠血糖、血脂代谢异常的同时,能降低大鼠肝脏IRS-2的表达,而运动能够增加大鼠肝脏IRS-2的表达,运动对血糖和血脂的改善作用可能与IRS-2的表达增加有关。 Objective To observe the insulin receptor substrate (IRS)-2 expression in the hepatic tissues of rats with hyperlipidemia following swimming exercise, so as to determine the role of IRS-2 in the regulation of lipid metabolism. Methods A total of 26 Wistar rats were divided into 3 groups: 9 in control group, 9 in high-fat feeding group and 8 in high-fat feeding plus swimming exercise group (exercise group). IRS-2 mRNA and protein expression was determined by RT-PCR and Western blotting assay. Results ① Serum triglycerides (TG), total cholesterol (TC) and low-density lipoproteins cholesterol(LDL-C) levels were elevated in high-fat feeding group compared with normal control group and exercise group(P〈0.05 or 0.01); high-density lipopro- teins cholesterol(HDL-C) level in exercise group was higher than that of the high-fat feeding group(P〈0.05). ② IRS-2 mRNA and protein expression was decreased in high-fat feeding group compared with that in the normal control group and exercise group (P〈0.01 or 0.05). Conclusion High-fat food can lead to abnormalities in blood glucose and lipid metabolism in rats; meanwhile, it can lower the expression of IRS-2 in the rat liver. Exercise can increase the expression of IRS-2, which may explain the association between improvement of lipid and glucose metabolisms and exercise.
