PPAR-γ配体在激素不敏感型前列腺癌中的作用

摘要 目的:研究过氧物酶体增殖因子活化受体(PPAR-γ)配体对人前列腺癌细胞系生长的影响。方法:用Northernblot、MTT法和流式细胞仪(AnnexinV和PI双染色)分别分析检测PPAR-γ配体15脱氧-△12,14-前列腺素J2(15-d-PGJ2)和曲列格酮(TGZ)对激素不敏感型的人前列腺癌细胞系PC-3细胞生长的影响。结果:PPAR-γ在前列腺癌细胞系PC-3高表达,其配体15-d-PGJ2和TGZ均以剂量依赖方式抑制细胞生长,使细胞生存率下降;用流式细胞仪(AnnexinV和PI双染色)分析结果为Annexin V-FITC阳性和PI阴性,提示15-d-PGJ2和TGZ能够促进前列腺癌细胞的早期凋亡。结论:PPAR-γ的配体可以通过激素不敏感型前列腺癌细胞上的受体,促进前列腺癌细胞的早期凋亡,使得细胞生存率降低,提示PPAR-γ可能成为治疗激素不敏感型前列腺癌的靶点。 Objective: To study the effects of proliferators - activated receptor-γ(PPAR -γ) ligand on growth of hormone - refractory human prostate cancer cells. Methods: After treated with 15 - deoxy - Δ^12,14- prostaglandin J (15 - d - PGJ2 ) and troglitazone(TGZ), respectively, the growth of hormone - refractory human prostate cancer PC - 3 cells were examined with the methods of Northern blot, MTT and flow cytometry(annexin V and propidium iodide staining). Results;PPAR-γ was strongly expressed in PC - 3 cells. Both 15 - d - PGJ2 and TGZ inhibited the PC - 3 cells growth in a dose - dependant way and the cell survival rate decreased. The results of flow cytometry were annexin - V - FITC positive and PI negative, which suggested that both 15 - d - PGJ2 and TGZ could induce PC - 3 cells apoptosis. Conclusion:PPAR-γ ligand can induce hormonerefractory human prostate cancer cell early apoptosis and decrease cell survival rate by inhibiting the receptor. This may be a new treatment of hormone - refractory human prostate cancer.
