摘要
首先对木霉菌T23在含有Mg、Cu、Zn、Fe、Mn、Mo、Ca共7种微量元素的组合培养基上经过7代继代诱导,然后运用DNA随机扩增多态性(RAPD)技术进行基因组DNA多态性分析。从70条引物中筛选出谱带清晰、重复性好的引物10条,共扩增出141条RAPD谱带,多态性谱带为107条,占扩增总谱带数的75.9%。DNA片段大小范围500~3000bp,并利用NTSYSpc2.1软件对扩增结果进行统计和聚类。结果表明:生防木霉菌T23诱变后具有一定的遗传稳定性和遗传多态性,生防木霉菌T23经过上述微量元素长时间诱导能发生遗传变异。
The biocontrol agent Trichoderma T23 and its induced strains sub-cultured for seven times on portfolio medium supplemented with 7 microelements including magnesium, copper, zinc, iron, manganese, molybdenum, and calcium, were subjected to a random amplified polymorphic DNAs (RAPD) study. Among 70 random primers tested, 10 distinct primers were selected for amplifying DNA fragments, in which, 75.9% of the 141 RAPD fragments showed polymorphism. The sizes of the DNA fragments were between 500bp and 3000bp. Furthermore,based on NTSYSpc2.1 cluster analysis that was conducted using strain T23 and the induced strains, the results showed that the induced strains of T23 was genetically stable and diverse. The variation was produced when T23 was induced by microelement for a long time period.
