摘要
以超级稻沈农9741的成熟胚为外植体,通过筛选基本培养基、优化激素组合和调节培养基渗透压,构建了适合水稻特定品种的高效再生体系。试验结果表明:愈伤组织诱导培养采用I3(NBB+2.0mg·L-1 2,4-D)培养基可获得95%的愈伤诱导率;愈伤组织继代培养采用S3(NBB+1.5 mg·L-12,4-D+0.2 mg·L-16-BA+500 mg·L-1脯氨酸)的培养基能使愈伤快速增殖,并且保持良好的胚性;愈伤组织分化培养采用D2(NBB+3.0 mg·L-1 6-BA+0.5 mg·L-1NAA+500 mg·L-1脯氨酸)的培养基可获得85%的幼苗分化率。该体系可用于超级稻沈农9741的遗传转化和基因工程育种研究中。
Using mature rice embryo from super rice Shennong 9741 as explants, a high efficient rice regeneration system suitable for special rice variety was established by selecting basic medium, optimizing the combinations of hormones and adjusting the osmotic pressures of the culture medium.Experimental results showed that the I3(NBB+2.0 mg·L^-1 2,4-D) induction medium was favorable for embryogenic callus induction, with the induction rate of 95%.The S3 (NBB+1.5 mg·L^-1 2,4-D+0.2 mg·L^-1 6-BA +500 mg·L^-1proline) subculture medium suited for maintenance and improvement of embryogenic calli, and the highest regeneration frequency reached 85 % in D2(NBB+3.0 mg·L^-1 6-BA +0.5mg·L^-1 NAA+500 mg·L^-1 proline) differentiation medium. The system established in this study could be in genetic manipulation and cellular engineering for the breeding of super rice Shennong 9741.
