茉莉叶石蜡制片技术的改进

Improvements on Paraffin Sectioning Technique for Jasminum sambac Leaf

以茉莉叶片为材料,对石蜡常规制片的部分步骤在时间上作了改进,并对制成的片子在光学显微镜下观察、拍照,以期寻找最佳观察结果和高效的制片方法。实验结果表明,以清洗3次(15 min/次)、脱水4次(30 min/次)、透明2次(30 min/次)、浸蜡2次(120 min/次)、脱蜡2次(30min/次)、番红染色50min的细胞结构完整,图像效果最佳,全程耗时624min,与常规石蜡制片的需要时间(2464 min)比较,可节省1840 min。清洗时间不低于15 min/次,脱水时间不低于30 min/次,对细胞结构和图像清晰度影响不大,但透明时间低于30 min/次、浸蜡时间低于120 min/次对细胞结构和图像清晰度有一定影响,若透明时间高于30 min/次、浸蜡时间高于120 min/次则浪费时间,而且造成组织块变硬变脆,影响制片质量。

The station time of part processing schedule on paraffm sectioning in Jasmimum sambac＇ leaves was optimized, the slides made from the optimized processing schedule based on orthogonal design were observed and photographed using light microscope to explore perfect observation results and least time-consuming methods in this paper. The result showed that cellular structure that was cleaned thrice （once for 15 min）, dehydrated four times （once for 30 min）, cleared twice （once for 30min）, infiltrated by paralTm twice （once for 120 min）, deparalTmizated twice （once for 30min）, and stained by safranine was intact and the best in image effect. The full process consumed 624min, whose paraffim sectioning period can, compared with regular one （2464 min）, save 1840min. cleaning time not less than 15min once and dehydration time not less than 30 min once have no effect on cellular structure and image defmition while cleating time less than 30min once and pamffm infiltrating time less than 120 min once have a certain effect on cellular structure and image definition, clearing time more than 30 min once and clearing time more than 120 min once lead to waste on station time, also hardening and crisping tissue group and worse sectioning quality.
