摘要
以人的浆液性卵巢上皮癌细胞系HO-8910为抗原,制备分泌抗卵巢癌单克隆抗体的杂交瘤,经间接免疫荧光组化鉴定证实。制备的单克隆抗体0C1C3与HO-8910呈强阳性反应,与其他肿瘤细胞系则无反应或反应极弱。在冰冻切片中,单克隆抗体OC1C3与卵巢恶性肿瘤呈强阳性反应(7/8),与绒癌有部分交叉反应(1/2),与正常卵巢和胚胎组织无反应。结果表明OC1C3是卵巢恶性肿瘤的一种特异性抗原的单克隆抗体。为进一步改善卵巢恶性肿瘤的免疫诊断方法提供了有利的基础。
The hybridoma secreting monoclonal antibody against human ovarian cancer was produced by immunization of BALB/C mouse with human ovarian epithelial cancer cell line HO-8910. Spleen cells of the immunized mouse were fused with mouse myeloma cell line SP2/0. Cell ELISA was used to select the positive cell colonies. After subcloning 4 times, cell stain(1C3) secreting monoclonal antibody with high specificity for ovarian cancer was collected. The isotype of the monoclonal antibody OC1C3 is IgG1 and its affinity constant is 2. 3 X 10 8. Reactivity of monoclonal antibody OC1C3 was investigated. The results showed that it has strong positive reactivity with malignant human ovarian tissues and ovarian cancer cell lines f while no reactivity with a number of other tumor tissues and fetal tissues. The epitope of OC1C3 recognized antigen is a new maker different from CA125 for ovarian epithelial cancer.
