摘要
本文应用斑点分子杂交(Dotblot)和斑点酶免疫测定法(DotEIA)对广州和海南地区3个鸭种携带鸭乙型肝炎病毒(DHBV)进行了调查,38只野水鸭血清标本DHBV自然感染率为63.2%(24/38).其一日龄雏鸭DHBVDNA阳性率为80%(8/10)。在国内外未见类似报导。应用野水鸭阳性DHBVDNA血清以0.02~0.2ml/只量感染8只1日龄DHBVDNA阴性的广州麻鸭,获得100%感染,且病毒血症至少可持续60天,该野水鸭经鉴定为驯养的野生绿头鸭种,此将为DHBV基因分析比较与建立一种慢性感染的鸭乙肝实验模型提供良好的线索。
The serological survey of the three kinds of ducks carried duck hepatitis B virus (DHBV) in Guangzhou and Hainan areas was undergone by methods of Dot Blot and Dot EIA. The study found that DHBV natural infection rate of 38 wild water ducks (Anas Platyrhynchos) Was 63. 2 % (24/38)by detecting DHBV DNA in serum samples.The DHBV DNA posiive rate of the wild water ducklings was 80% (8/10 ). 8Guangzhou ducklings with negative result of DHBV DNA test were injected intravenously with the posltive DHBV DNAserum from the wild water ducks (at doses of 0.02-0. 2ml per duck). It led to 100% of infection rate of the 8 Guangzhou ducklings, and the virusemia maintained for at least over 60 days. The wild water duck was identified as a specie of wild green head duck and may be used as comparison research of DHBV gene and as an anirnal experiment model of DHBV chronic infection.
