摘要
介绍一种用毛细管柱气相层析直接测量红细胞膜脂肪酸总组分的新方法。首先用梯度离心从全血中分离出红细胞,提取细胞膜,用无水盐酸甲醇一步法处理提取脂肪酸甲基酯,经过气相层析,直接测出红细胞膜总脂肪酸成分。结果表明:该方法稳定可靠,可重复性强,精确度及分辨率高,方法简便易行,可减少实验时间,为红细胞膜研究开辟了一条途径。
A new method to analyse the total fatty acid composition of erythrocyte membrane with capillary gaschromatography is introduced.First,erythrocyte cells are separated from the whole blood by gradient centrifugation.After hemolysis,the erythrocyte membranes are extracted by lowtemperature centrifugation.Finally,with onestepmethod the fatty acids are methylized with 3N waterfree HClMethanol.Our experiments indicate that this method is stable,reliable,and repeatable.It has high precision and identification.This method is also simple to be handed and timesaving.
