摘要
目的:建立散寒解热颗粒剂中葛根素的含量测定方法。方法:样品用甲醇超声提取,采用高效液相色谱法,Nuc leosil-C18(200mm×4.6mm,7μm)分析柱,乙腈∶水(内含0.02moL/L磷酸二氢钠用10%磷酸调pH值为4.0)(12.5∶87.5)为流动相,检测波长265nm。结果:葛根素峰的保留时间约为13min。并与其邻近峰的分离度均大于1.5,葛根素的线性范围为0.4-1.4μg(r=0.9999),平均回收率为98.7%(n=5)。结论:方法简便,快速,结果准确可靠,可作为散寒解热颗粒剂中葛根素的含量测定方法。
Objective:To establish a method for determining the concentration of puerarin in Sanhanjiere granules.Methods:The sample was extracted with methanol on a supersonic.HPLC was used for the determination of puerarin in Sanhanjiere granules.The chromatographic procedure was carried out using Nucleosil-C18(200mm×4.6mm,7μm) as an analytical column and a mixture of 12.5 volume of acetonitrile,87.5 volume of water which was contained 0.02moL/L Sodium dihydrogen phosphate and adjusted the solution pH4.0 by phosphoric acid as mobile phase.The detection wavelength was set at 265nm.Results: The peaks of puerarin appeared on about 13 minutes and the resolutions of them were more than 1.5.Their linear ranges were between 0.4 and 1.4g(r=0.9999),with the average recoveries of 98.7%(n=5).Conclusion:This method is sensitive,quick and accurate for the measurement of puerarin in Sanhanjiere granules.
