从制革废灰碱牛皮中提取天然胶原(Ⅱ)——胶原的物理化学性能表征 被引量:4

摘要 对经EDTA和盐酸预处理后提取得到的天然胶原的物理化学性能进行了表征。经盐酸预处理后提取得到的胶原(HPC)的等电点(5.38±0.02),低于经EDTA预处理后提取得到的胶原(EPC)的等电点(5.87±0.03),HPC的自由氨基含量低于EPC;同时,HPC的平均粒径和相对黏度均大于EPC。两者的氨基酸组成相似,与商品小牛皮胶原(CCC)差别不大;通过电泳、圆二色谱分析分别验证了EPC和HPC的多肽链和三股螺旋结构的完整性。差式扫描量热法分析表明:EPC和HPC与商品小牛皮胶原一样,具有较好的热稳定性。 The physicochemical properties of collagen extracted from EDTA - and HCl - pretreated bovine limed split wastes respectively were charactered. The isoelectric point of HPC ( 5.38 ± 0.02) is lower than that of EPC (5.87 ± 0.03 ). And the primary amino acid content of HPC is lower than that of EPC. Simultaneity, HPC has larger Z - particle size and higher relatively viscosity than those of EPC. The amino acid compositions of EPC and HPC are similar to each other, although there is a little difference compared to that of commercial calfskin collagen (CCC). The results of sodium dodecyl sulphate polyacrylamide gel electrophoresis analysis (SDS -PAGE) and circular dichroism (CD) spectras represented the integralities of their polypeptide and triple helix conformations. Thermal behaviors of EPC and HPC, reviewed using differential scanning calorimetry ( DSC), are almost as same as that of CCC.
