海藻萜类化合物对大鼠急性酒精性肝损伤的保护作用研究 被引量:9

Prevention and Treatment of Acute Alcoholically Hepatic Injury with Laurencia Extract in Rats

摘要 目的:观察凹顶藻萜类化合物(Laurencia extract,LET)对大鼠急性酒精性肝损伤的保护作用,并探讨其可能的作用机制。方法:雄性Wistar大鼠60只,随机分为6组。A组为空白对照组,每日给予蒸馏水18mL/(kg·d)灌胃。B组为酒精模型组,每日给予乙醇7.2g/(kg·d)灌胃,乙醇体积分数为50%。C、D、E组分别为LET低、中、高剂量干预组,每日分别给予LET25、50、100mg/(kg·d)灌胃。F组为甘利欣药物对照组,每日给予甘利欣200mg/(kg·d)灌胃。C、D、E、F组酒精剂量同模型组。试验进行了7d。末次灌胃24h后,乌拉坦麻醉,腹主动脉取血,分离血清,留取肝组织。用电镜观察肝组织超微结构的变化。检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、甘油三酯(TG)、胆固醇(CHO)水平。结果:电镜下观察到B组肝细胞内线粒体肿胀,脂滴数量增加,Kupffer细胞周围可见胶原纤维;C、D组线粒体结构基本正常,部分肝细胞胞浆内可见少量脂滴;E组肝细胞内线粒体数目增多;F组肝细胞内脂滴数量较多,狄氏隙内偶见胶原纤维。B组大鼠血清ALT、AST、TG、CHO均高于空白对照组,差异具有统计学意义(P<0.05);而不同剂量的LET干预组和酒精模型对照组比较,血清ALT、AST、TG、CHO水平均有不同程度地降低,其中LET中、高剂量干预组血清ALT、AST、TG、CHO水平明显低于模型组,差异具有统计学意义(P<0.05)。结论:LET可明显改善肝组织超微结构的病理损伤,可降低因酒精暴露而引起的血清转氨酶增高,降低机体血脂水平,对酒精造成的脂质代谢紊乱有一定的调节作用。 Objective: The protective and therapeutic effects of Laureneia extract (LET) on acute alcoholically hepatic injury in rats was evaluated in this paper. Methods: 60 male Wistar ruts were randomLy assigned to one of the six groups: Group A (placebo group) was orally given with distilled water of 18 mL/(kg·d); Group B was drink with 50% alcohol of 7.2 g/(kg·d); Group C, D and E was orally supplemented with LET of 25, 50 and 100 mg/(kg·d) respectively; Group F (positive control) was fed with diammonium glycyrrhizinate of 200 mg/(kg·d) . All rats were treated for 7 days. At the 8th day of the treatment, the rats blood were collected to measure ALT, AST, TG and CHO. The ultrastructures were studied. Results: Compared with control group, Group B showed severe hepatic injury with abnormally higher ALT, AST, TG and CHO (P〈0.05), as well as hepatocyte fatty degeneration. The levels of ALT, AST, TG and CHO in blood serum in the 50, 100 mg/kg supplement groups were lower than the model group (P〈0.05) . LET and diammonium glycyrrhizinate can decrease the levels of ALT, AST, TG and CHO in the blood tests and hepatic injury induced by alcohol. Conclusion: LET may prevent rats from acute alcohol-induced hepatic injury and favorable regulating effect on dyslipidemia provoked by high ethanol.
