摘要
目的建立测定雷帕霉素在组织中含量的高效液相色谱方法。方法采用AlltechC18柱,以乙腈∶四氢呋喃∶(55∶40)为流动相,检测波长为278nm。结果线性回归方程为A=3.2481C+7.4082×10-水5∶3,r=0.9925。在4~128ng/ml范围内线性良好。结论本法操作简单,重现性好,可用于测定组织中雷帕霉素的含量。
Objective To establish a HPLC method for content determination of rapamycin in tissues. Method Alhech C18 column was used. The mobile phase was the mixture (acetonitrile:tetrahydro-furan: waler =55 :5 :40). The deteclion wavelength was 278nm. Results Calibration curves was A=3,2481C+ 7.4082×10^-3, r=0.9925. A good linearity was shown over the concentration range of 4-128ng/ml. Conclusion The method is simple and reproducible. It is suilable for content determination of rapamyein in tissues.
