IL-1β干预后人牙周膜成纤维细胞中TRAF6的基因表达研究

摘要 目的:研究IL-1β干预后人牙周膜成纤维细胞中TRAF6的基因表达水平的变化,为细胞因子网络调控牙周膜成纤维细胞功能的研究提供新资料。方法:采用原代培养的人牙周膜成纤维细胞,取5~8代细胞以IL-1β梯度浓度干预,采用RT-PCR法检测TRAF6在人牙周膜成纤维细胞中的表达。结果:在正常的人牙周膜成纤维细胞中未见TRAF6基因表达,IL-1β干预后,TRAF6呈阳性表达,并且随着IL-1β干预的浓度增高,其表达水平上调。结论:IL-1β干预后TRAF6在HPLFs中的表达水平的变化提示TRAF6可能是参与牙周膜成纤维细胞功能调控的重要信号分子。 Objective:To examine the expression of tumor necrosis factor receptor-associated factor-6 (TRAF6) in human periodontal ligament fibroblasts (HPLFs) by the interference of IL-1β, and provide new document to the research of the effect of cytokine network to periodontal ligament fibroblasts. Method:To isolate and culture HPLFs, and make biological assay. IL-1β interfered the cells of the fifth to eighth passage by various concentrations,then examined the expression of TRAF6 in HPLFs by RT-PCR method. Result:Through RT-PCR electropherogram and analysis,we didn't found TRAF6 gene band in normal control HPLFs group,but we found 216bp TRAF6 gene band,which display dependability to the concentration of IL-1β,express in HPLFs groups which affected by IL-1β. Conclusion:The results intimated that TRAF6 maybe a new found important signal factor which participates in the regulation of HPLFs' function and pefiodontitis.
