摘要
目的:制备容易保存的以滤纸为载体的HIV抗体质控品,研究不同洗脱液的洗脱效果及稳定性。方法:取相同的质控品原液制备10μl体系的液态型质控品和滤纸型质控品。液态型质控品使用时加入490μl正常人血清,滤纸型质控品分别用生理盐水组、血清组、PBS组、PBS-TW20组和样品稀释液组等不同稀释液进行溶解,对其质控测定值和稳定性进行研究。结果:5组不同稀释液溶解的滤纸型质控品检测结果与传统的液态质控品相比均无显著性差异(P>0.05),但生理盐水组和PBS组所得数值与液体型最符合,并且在1周内具有较好的稳定性和均一性。结论:滤纸型质控品可替代液态质控品用于HIV初筛实验室的室内或室间质控。采用容易获得的生理盐水作为滤纸型质控品的洗脱液,具有较好的稳定性和准确性。
Objective: To prepare filter - style HIV antibody specimens for internal quality control using dried plasma spots and investigate the changes of different liquid used in the fiher antibody washing. Methods:Pipette 10 μl original HIV positive plasma for internal quality control to mix with 490 μl normal serum as liquid - style control and same volume of HIV positive plasma is used in fiher paper control preparation. The stability of five group of liquid has been studied as filter paper elution of normal saline (NS), serum, PBS, PBS -TW20 and specimen dilution attached in HIV ELISA Kit. All data was controlled by ELISA Kit. Results :Though there is no significant difference among five elution washer used in fiher - style HIV positive control, NS and PBS have good stability and reduplication. Conclusion: HIV filterstyle control washed in NS and used in internal quality control test has good stability and reduplication, can replace traditional liquid control in clinic Lab.
