非小细胞肺癌中Bcl-xL的研究

摘要 目的:观察非小细胞肺癌组织与癌旁组织中Bcl-xL的基因转录和蛋白表达水平,探讨Bcl-xL在非小细胞肺癌发生发展中的可能作用,为非小细胞肺癌的诊断和治疗提供参考。方法:反转录PCR和免疫印迹的方法分别检测60例临床确诊的非小细胞肺癌组织与癌旁组织中bcl-xL基因表达和Bcl-xL蛋白的水平,并与肺良性病变组织比较。结果:非小细胞肺癌组织的bcl-xL mRNA水平明显高于癌旁组织和肺良性病变组织(P<0.01),mRNA水平与临床分期呈正相关,与其他临床病理相关因素(性别、年龄、肿瘤大小、发生部位、组织学类型、分化程度)无明显相关,而癌旁组织和肺良性病变组织bcl-xL mRNA水平之间没有差别(P>0.05)。蛋白表达检测结果与mRNA水平高度一致。结论:非小细胞肺癌组织中bcl-xL基因和蛋白表达水平明显升高,可能促进了肿瘤的发生发展,可以考虑作为非小细胞肺癌临床分期和评价临床预后的参考指标。 Objective:To observe the expression of bcl - xL gene and Bcl - xL protein in non - small - cell lung cancer tissues and adjacent tissues, and to study Bcl - xL role in the development and progression of non - small - cell lung cancer. Methods: The mRNA levels of bcl - xL were detected by RT - PCR with its corresponding primers, and its protein expression levels were detected by Western Blot with mouse anti - Bcl - xL monoclonal antibody in 60 clinical confirmed non - small - cell lung cancer tissues and their corresponding adjacent tissues ( more than 5 cm far away from the tumor tissue) as well as benign lung lesion tissues from non - cancer patients. Results: The mRNA and protein levels of Bcl - xL were much higher in cancer tissues than adjacent tissues and benign lesion tissues ( P 〈 0. 01 ), and expression level was positively correlated with clinical stages of the cancer (P 〈 0. 05 ), and was not correlated with other factors such as sex, age, tumor size, tumor location and morphological types. Well, no difference detected between adjacent tissues and benign lesion tissues (P 〉 0. 05). Conclusion:Bcl -xL expression is obviously highly positive correlated with stages, which suggested that Bcl -xL may enhance the development and progression of non - small - cell lung cancer and may could be a prognostic indicator for this kind of tumor.
