摘要
在温度为25℃,处理时间为6h、12h、24h、48h的条件下,用浓度为0.05%、0.1%、0.2%、0.5%的秋水仙素对二倍体的菘蓝萌发种子进行诱导,并对诱导后的植株幼苗在分蘖期进行细胞学和形态学的研究。结果表明:用0.05%~0.5%浓度的秋水仙素处理的萌发种子均可以获得四倍体植株,0.2%秋水仙素溶液处理6h的诱导加倍的成功率最高,加倍植株均为二倍体和四倍体的混倍体;混倍体植株叶片气孔保卫细胞的长度和气孔的长度均高于未加倍的二倍体植株。
Doubled Isatis indigotica Fort. germinating seeds were induced by Colchicine which concentration were 0.05 %,0.1% ,0.2 % and 0.5 % under 25℃, and the treatment time were at 6 hour, 12 hour,24 hour and 48 hour, the cytology and morphology of the induced seedling at tillering stage were studied. The result showed that: gemmiferous seeds treated with 0.05 %-0.5 % aqueous solution of colchicines could get the polyploidy plants, the most suitable concentration and treatment time were 0.2 % and 6 hour, all doubled plants were mixoploids including digploids and tetraploids. The leaves stoma of defending cells and stoma were longer than that of diploids.
