摘要 以猪心脏为原料,确定总磷脂提取的优化条件。设计了以氯仿-甲醇混合溶剂为提取剂的提取工艺试验,探讨了氯仿-甲醇的比例、液料比、水洗比例、提取时间对总磷脂含磷量的影响。并采用二次回归正交旋转组合设计方法,对总磷脂的提取条件进行优化。结果表明,最佳提取条件为:氯仿-甲醇的比例2∶1、液料比22∶1、水洗比例27%、提取时间5h,总磷脂含磷量为20.45mg·mL-1。 Making pig hearts as the material to dtermine the optimum technological condition on extraction of total phospholipids. Chlo- roform-methanol was adopted for extraction, the effect of chloroform to methanol ratio, liquid to material ratio, extraction time and vol- ume percentage of water in washing were investigated. The technological condition was optimized by quadratic regress orthogonal rotary combination design. The results indicated that the optimization condition was as followed: chloroform to methanol ratio was 2:1, liquid to material ratio was 22:1, volume percentage of water in washing was 27 %, extraction time was 5 hours and the phosphorus content was 20.45 mg·mL-1.
