摘要 目的:建立高效液相色谱-质谱法定量测定中药材浙贝母中贝母素甲、贝母素乙的含量方法。方法:采用ZORBAX Eclipse XDB-C_(18)(150 mm×2.1 mm,5μm),以甲醇-10 mmol·L^(-1)醋酸铵溶液(40:60)为流动相,流速为0.2 ml·min^(-1),用质谱检测器检测。结果:贝母素甲乙的线性范围分别为0.94~28.13 mg·L^(-1)(r=0.997)和0.95~28.37 mg·L^(-1)(r=0.998 0),平均回收率分别为105.3%(RSD4.6%),106.4%(RSD4.8%)(n=6)。结论:方法灵敏、准确,非常适用于检测含量低,无紫外吸收的化学成分。 Objective: To establish an HPLC-MS method for determination the contents of peimine and peiminine in Thumberg Fritillary Bulb. Method : The Zorbax Eclipse XDB-C18 column( 150 mm× 4.6 mm, 5 μm) was used, the methanol-10 mmol· L^ -1 ammonium acetate solution(40: 60) as mobile phose, the flow rate was 0.2 ml·min^-1 with MS detected. Result: The linear ranges of pei- mine and peiminine were at 0.94 -28.13 mg·L^-1 (r =0.997) and 0.95 -28.37 mg·L^-1 (r =0. 998 0), the average recoveries (r = 6) were 105.3% and 106.4% with RSD 4.6% and 4.8% respectively. Conclusion: The method is sensititive and accurate, it can be used for determination of chemical constituents with low content and without ultraviolet absorption.
