摘要
本试验采用LPS刺激小鼠巨噬细胞RAW264.7细胞0、6、9、12、18、24h,观察不同种类β-防御素(mouse beta defensins,mBDs)mRNA表达水平的影响;采用含有0.2、0.4、0.6、1.0mg/mL苜草素的细胞培养液处理RAW264.7细胞,观察其对RAW264.7细胞mBDs的mRNA表达水平的影响。结果表明:mBD-1和mBD-4的表达不受LPS影响,mBD-2和mBD-3的表达受到LPS诱导时间的影响;含苜草素的细胞培养液可使mBDs的表达上调,0.2mg/mL效果最明显。苜草素可以使RAW264.7细胞mBDs的表达升高,这可能是苜草素参与免疫调节,提高动物机体免疫力的机制之一。
To investigate the mechanism of polysavone improving the immunity of animal body,the mouse macrophage line RAW264.7 cells were stimulated by LPS at 0,6,9,12,18,24 h respectively,and the mRNA expression of mouse beta defensins (mBDs)was detected.Polysavone with different dose were used to culture RAW264.7 cells,and the mRNA ex- pression of different kinds of mBDs (mBD-1 - 4)were detected.The results showed:The mBD-2 and mBD-3 mRNA expression could be improved by LPS,and it is related to the culturing time,but there were no effects on the mRNA expression of mBD-1 and mBD-4.The medium containing polysavone could improve the mRNA expression of mBDs,especially the treatment with 0.2 mg/mL polysavone.Polysavone could improve the mRNA expression of mBDs in RAW264.7,that might be one of the reasons which polysavone improve the animal immunity.
