摘要
目的:从肝细胞线粒体DNA(mtDNA)和氧自由基的角度,探讨六味抗衰饮延缓衰老的可行性和可能的作用机制,为临床提供实验依据.方法:将36只SD大鼠随机分成3组,每组12只,即青年对照组、衰老模型组、六味抗衰饮组.衰老模型组、六味抗衰饮组给予D-半乳糖溶液造模,青年对照组给予生理盐水.六味抗衰饮组在造模的同时给予治疗,青年对照组和衰老模型组给予等体积生理盐水灌胃,连续40d.检测肝细胞mtDNA的相对含量、血清SOD活性及MDA含量作比较.结果:六味抗衰饮能够降低衰老状态下大鼠肝细胞mtDNA相对含量及血清MDA含量,提高血清SOD活性(P<0.05).结论:六味抗衰饮组具有减轻D半乳糖诱导亚急性衰老动物模型的氧化损伤,保护肝细胞mtDNA,从多方面延缓机体衰老的作用.
AIM: To study the feasibility and possible mechanism of anti-aging of Liu Wei Kang Shuai Yin from the aspects of hepatocyte mitochondrial DNA (mtDNA) and oxygen free radicals, thus to provide the experimental evidence for its clinical application. METHODS: Thirty-six SD rats were randomly divided into 3 groups, young control group, aging model group, and Liu Wei Kang Shuai Yin group, with 12 in each group. The D-galactose was given to aging model group and Liu Wei Kang Shuai Yin group to establish the aging models, while normal saline to young control group. As for Liu Wei Kang Shuai Yin group, Liu Wei Kang Shuai Yin was administrated when the aging models were established, while normal saline to aging model group and young control group, which lasted for 40 days. Then, the hepatocyte mtDNA content, SOD activity and MDA content in serum were analyzed and compared. RESULTS: Liu Wei Kang Shuai Yin reduced the hepatocyte mtDNA content and serum MDA content and promoted the serum SOD activity in the aging rats ( P 〈 0.05 ). CONCLUSION: Liu Wei Kang Shuai Yin can relieve the oxidative damage in D-galactose induced aging animal model, protect hepatocyte mtDNA, therefore relay body aging from several aspects.
