银杏叶提取物EGB761对肢体制动大鼠骨骼肌萎缩的防护效应 被引量:1

摘要 目的:探讨银杏叶提取物EGB761对废用性骨骼肌萎缩的防治作用.方法:选用雄性SD大鼠24只,随机分为空白组,对照组,试验组,每组8只.大鼠右后肢用可塑性石膏由足跖管型固定至膝上1cm处,膝关节固定于屈曲位100°,踝关节固定于跖屈60°,共4wk.对照组每日给予蒸馏水3mL灌胃,试验组每日给予银杏叶提取物EGB761水溶液灌胃(灌胃剂量120mg/kg,浓度14g/L).4wk后完整解剖右后肢腓肠肌,检测腓肠肌收缩张力、肌湿重、蛋白质含量、Na+-K+-ATP酶及Ca2+-ATP酶活性.结果:对照组、试验组与空白组相比,腓肠肌收缩张力、肌湿重、蛋白质含量、Na+-K+-ATP酶及Ca2+-ATP酶活性显著下降(均P<0.05).试验组与对照组相比,试验组的上述指标显著高于对照组(均P<0.05).结论:银杏叶提取物EGb761能够显著提高废用性萎缩骨骼肌的收缩张力、肌湿重、蛋白质含量、Na+-K+-ATP酶及Ca2+-ATP酶活性,从而有效的抑制骨骼肌废用性萎缩的发生. AIM : To study the preventive and therapeutic effects of ginkgo biloba extract 761 on disuse atrophy of skeletal muscles. METHODS: Twenty-four SD rats were divided into blank group, control group and experimental group, with 8 rats in each group. The right hind limbs of the rats were fixed for 4 weeks with compliant plaster from metatarsus to 1 cm upper knees, and the knee joints were fixed in 100° and ankle joint in 60° extension position. The rats in control group received administration of distilled water (3 mL) , and the rats in experimental group received administration of ginkgo biloba extract 761 ( 120 mg/kg, 14 g/L). After 4 weeks, gastroenemlus muscles in right hind llmb were dissected and measured for the contractility, wet weight, protein content, Na^+ -K^+ -ATPase and Ca^2+ -ATPase activities. RESULTS : Compared with blank group, contractility, wet weight, protein content, Na^+-K^+ -ATPase and Ca^2+ -ATPase activities of gastroenemius muscles in control and experimental groups significantly decreased (P 〈 0.05 ). Compared with control group, the above parameters of gastroenemius muscle were obviously improved in experimental group ( P 〈 0.05 ). CONCLUSION : Ginkgo biloba extract 761 can improve contractility, wet weight, protein content, Na^+ -K^+ -ATPase and Ca^2+ -ATPase activities in disuse atrophy of skeletal muscles, suggesting that Ginkgo biloba extract 761 has preventive and protective effects on development of disuse atrophy of skeletal muscles.
