摘要
通过碳源和氮源的单因素实验,以及采用葡萄糖、黄豆粉、KH2PO4和MgSO4.7H2O为实验因素的L16(45)正交实验,确定了裂褶菌半合成发酵培养基的最优配方为:葡萄糖50.0g/L、黄豆粉6.0g/L、KH2PO43.0g/L、MgSO4.7H2O0.5g/L;在5L发酵罐的培养研究中发现,裂褶菌在半合成培养基中培养时,菌体量和胞外多糖量比在谷物汁天然培养基中培养时分别高出17.2%和30.0%,说明半合成培养基更利于裂褶菌生长和胞外多糖的分泌。半合成培养基成分简单、易于配制、成本较低,对裂褶菌的工业化培养具有实际应用价值。
The optimization of semisynthetic medium for Schizophyllum commune was studied. The medium components were optimized by both single factor experiment and orthogonal experiments, and the optimal medium contained: glucose 50.0g/L, soybean powder 6.0g/L, KH2PO4 3.0g/L, MgSO4 · 7H2O 0.5g/L. In the study of Schizophyllum commune cultivated in a 5L fermentor, when semisynthetic medium was used, the yield of mycelium and production of exopolysaccharide were 17.2% and 30.0%, which was higher than that in the wort medium, respectively. This low cost semisynthetic medium was easier to make, so it is more applicable than the wort medium in the industrial culture of Schizophyllum commune.
