摘要
以柞蚕蛹为原料,采用索式提取法,以石油醚、丙酮和环己烷为溶剂进行脱脂,然后以1%NaOH和0.9%NaCl乙醇溶液提取脱脂柞蚕蛹中的碱溶和醇溶蛋白,同时采用SDS-PAGE凝胶电泳进行分析。实验发现,碱溶蛋白提取率高于醇溶蛋白。电泳分析结果为,碱溶蛋白8条带为:178、72、64、62、42、30、28、16kDa;醇溶蛋白7条带为:178、64、62、42、30、28、16kDa。
Using Soxhlet extraction to defat oak silkworm with petroleum ether, acetone, cyclohexane. Then, the protein content of samples with 1% NaOH and 0.9% NaCl were determined. At last, SDS-PAGE was used to analyze furthermore. The study indicated alkali extracted protein was prior to ethano. In SDS-PAGE, alkali extracted protein showed 8 bands were: 178, 72, 64,62,42,30,28, 16kDa; ethano extracted protein showed 7 bands were : 178.64,62.42.30.28,16kDa.
