摘要
目的:探讨巨细胞病毒感染对小鼠学习记忆的影响、Ca2+/钙调蛋白依赖性蛋白激酶Ⅱ(CaMKⅡ)的表达,及其在学习与记忆中的作用。方法:采用颅内注射制造巨细胞病毒颅内感染小鼠模型,利用Morris水迷宫试验对病毒感染组和对照组动物的学习和记忆成绩进行3 d测试,记录其学习和记忆成绩,同时观察海马组织神经细胞[Ca2+]i及CaMKⅡ在胞膜中表达水平的变化。结果:Morris水迷宫测试的结果显示,病毒感染组小鼠的学习记忆成绩明显下降,神经细胞[Ca2+]i较对照组增高,胞膜蛋白CaMKⅡ的水平表达下降,且差异有显著性(P<0.01)。结论:巨细胞病毒感染能够导致小鼠学习记忆能力下降及CaMKⅡ在神经细胞胞膜的表达下降,说明CaMKⅡ参与了巨细胞病毒对学习记忆的影响。
Objective To explore the effect of CMV infection on murine learning and memory and influence of the expression of calcium/calmodulin-dependent protein kinase Ⅱ (CaMKⅡ)in mice hippocampus. Methods CMV- infected mice model were prepared by intracerebral inoculation with CMV suspension. The control were injected with sodium chloride. The performance of spatial learning and memory were measured with Morris Water Maze for 3 days respectively, meanwhile, CaMK Ⅱ expressions in hippocampal were observed. Results The behavior and responses of the CMV-infected group were much slower than control group in learning and memory (P 〈 0.01 ). The CaMK Ⅱ expressions on the neurocytic membrane were significantly decreased than that of control group (P 〈 0.01 ). Conclusions The capacity of learning and memory and the expression of CaMK Ⅱ are decreased in the CMV-infected group. Our results suggests that the CaMK Ⅱ may participate in the effect of CMV infection on learning and memory.
