摘要
目的研究大鼠灌胃棉花皮素-8-O-葡萄糖醛酸苷后血清中药物的分析检测方法。方法大鼠灌胃250mg/kg棉花皮素-8-O-葡萄糖醛酸苷,收集血样,进行适当的前处理,采用HPLC-DAD、LC-MSn法对血清样品进行检测。结果HPLC-DAD和LC-MSn两种方法均能检测到血清中的棉花皮素-8-O-葡萄糖醛酸苷,但未能检测到其代谢产物。血清样品前处理中的离心转速、pH值、纯化方法对血清中棉花皮素-8-O-葡萄糖醛酸苷的分析检测有明显影响。结论建立了大鼠灌胃棉花皮素-8-O-葡萄糖醛酸苷后血清中药物的快速灵敏的分析检测方法,为棉花皮素-8-O-葡萄糖醛酸苷的进一步药动学研究提供参考依据。
Objective To develop an analysis method used for rat serum after ig administration of gossypetin-8-O-glucuronide. Methods Rats were ig administrated by gossypetin-8-O-glucuronide at the dose of 250 mg/kg, then the blood was collected, pre-treated appropriately, and detected by HPLC- DAD and LC-MS^n. Results In rat serum, gossypetin-8-O-glucuronide was detected by both HPLC-DAD and LC-MS^n methods, but no other metabolites were detected by the both methods. The pre-treatment of samples, such as centrifugal speed, pH value, purification method, could markedly affect the detection of gossypetin-8-O-glucuronide in rat serum. Conclusion A rapid and sensitive method for analysis of rat serum after ig administration of gossypetin-8-O-glucuronide is established and this study will be useful for the further study of gossypetin-8-O-glucuronide.
