邻苯二甲酸二丁酯对EL4细胞增殖作用的影响

摘要 目的探讨邻苯二甲酸二丁酯(DBP)对激活与未激活的小鼠淋巴瘤细胞(EL4细胞)增殖作用的影响。方法用佛波醇酯(简称PMA,1、5、10ng/m1)激活EL4细胞,分别用不同浓度的DBP(0.1、1、2.5、5、10μmol/L)在无或有PMA激活条件下作用EL4细胞24h及48h,以MTT法检测细胞增殖情况。结果与对照组相比,不同浓度的DBP单独作用于未激活的EL4细胞24h及48h均未引起细胞明显增殖。以5ng/ml PMA激活EL4细胞,0.1~2.5μmol/LDBP作用细胞24h就能明显促进细胞增殖,而5μmol/LDBP在48h时细胞增殖作用显著(P〈0.05)。结论DBP能明显促进激活的EL4细胞增殖,且DBP与PMA对EL4细胞增殖有协同作用,可能与DBP的免疫毒性有关。 Objective To explore the effects of dibutyl-phthalate(DBP)on the proliferation of active and inactive EL4 cell. Methods Phorbol-12-myristate-13-acetate(PMA)(1, 5 and 10 ng/ml)was used to activate EL4 cell, and EL4 cell was exposed to 0.1, 1, 2.5, 5 and 10 μmol/L DBP with or without PMA for 24 h or 48 h respectively. The proliferation of EL4 cell was analyzed by MTT assay. Results Compared with the control, DBP alone did not enhance the inactive EL4 cell's proliferation significantly after 24 h or 48 h. When EL4 cell was activated by 5 ng/ml PMA, 0.1 - 2.5 μmol/L DBP significantly increased the proliferation of EL4 cell after 24 h, and 5 μmol/L DBP significantly enhance the proliferation of EL4 cell after 48 h. Conclusions DBP could significantly enhance the proliferation of the active EL4 cell, and there was synergistic effect on the proliferation of EL4 cell between DBP and PMA, which might be relevant to the immunotoxicity of DBP.
