摘要
目的探索分解卵巢黏液性癌表面黏液的方法及其对化疗药物泰素作用于人卵巢黏液性癌细胞株OMC685的影响。方法2007年4月至11月于复旦大学妇产科研究所运用免疫细胞化学法分析OMC685所分泌黏液中粘蛋白的类型,通过查阅文献筛选出适当的黏蛋白分解酶,用ELISA法证明黏液的分解,并于黏液分解后加用泰素,MTT法检测OMC685增殖的变化。选取人卵巢浆液性癌细胞株SKOV3加入含黏蛋白培养液后,再给予泰素,MTT法检测细胞增殖。结果OMC685表面所包绕黏液成分为黏蛋白2(mucin2)。α-N-内切乙酰半乳糖苷酶能够较特异地分解黏液。黏液分解后加用泰素对OMC685细胞增殖的抑制显著增强,而SKOV3加含粘蛋白的培养液后再加入泰素,对细胞增殖的抑制作用显著减弱。结论卵巢黏液性癌细胞分泌的黏液可能起到一定的屏障保护作用,通过使用分解酶分解黏液能够提高化疗药物泰素的作用效果。
Objective To explore the anti-cancer effect of the chemical agent taxol on an ovarian mucinous cancer cell line OMC 685 after the outside mucous layer of cellular surface is decomposed. Methods In April to November of 2007 at the Obstetrics and Gynecology Research Institute of the Fudan University, the main type of mucin within the mucous layer was analyzed by immunohistochemistry. Several proper mucinases were recognized through reviewing literatures, and the efficacy of each selected mucinase was certified by ELISA. The proliferation changes of OMC 685 were observed through MTT method. Compared with it, we selected ovarian serous carcinoma cell line SKOV3 to use taxol after covering complete growth medium containing mucin. Results Mucin 2 was the main protein type of mucin here. Endo-α-N-acetylgalactosaminidase specifically decomposed the mucous protein, and degraded the surface mucous layer. MTr method could further demonstrate that the proliferation- inhibiting effect of taxol was obviously enhanced by mucinase treatment, and SKOV3 was obviously reducod after covering medium containing mucin. Conclusion The mucous layer on the ovarian mucous cancer cell may act as a natural barrier against the anti-cancer chemical agents. The mucinase treatment is able to break down the mucous barrier, and then enhance the cytotoxic efficacy of taxol on OMC685 cancer cells.
