摘要
目的比较雪旺细胞在不同可降解组织工程支架材料上体外培养的生长活性。方法选择bFGF胶原-壳聚糖海绵、可吸收明胶海绵,和PLGA三种可降解支架材料,把原代培养的雪旺细胞悬液接种在支架材料上,用MTT法测定细胞增殖能力和生长情况;流式细胞仪测定细胞周期和细胞凋亡;环境扫描电镜观察细胞形态和材料结构,以此来评价雪旺细胞的生长活性。结果雪旺细胞可在bFGF胶原-壳聚糖海绵、可吸收明胶海绵,和PLGA等支架材料上生长,但与bFGF胶原-壳聚糖海绵相容性最好。结论bFGF胶原-壳聚糖海绵,更适合构建具有雪旺细胞活性的组织工程化人工神经支架材料。
Objective To compare the activity of Schwann cells cultured in different scaffold of artificial degradation in vitro. Methods To choose bFGF Chitosan - collagen Sponge, absorbable gelatin sponge and PLGA degradable supporting materials. Primary cultured Schwann cells were used to seed on supporting materials. Then, the Schwann cells of rat were isolated and cultured by sequential enzyme digestion . MTT assay was employed to observe the proliferation of Schwann cells, Determination of cell cycle and apoptosis by flowcytometer, shape of cells and structure of materials observed by environment scanning electron microscope, to judge the activity of Schwann cells. Results The Schwann cells may be cultured on bFGF Chitosan -collagen sponge, absorbable gelatin sponge and PL- GA, but best for bFGF Chitosan - collagen Sponge. Conclusion The bFGF Chitosan - collagen Sponge seeded with Schwann cells were suitable to construct tissue- engineered bioartificial.nerve.
