摘要
目的目前烧伤瘢痕机理尚不完全清楚,本研究旨在通过对血清中蛋白质组的质谱分析来阐述烧伤瘢痕血清蛋白质组的改变。方法随机取浅度烧伤患者40例(Ⅰ度烧伤20例,浅Ⅱ度烧伤20例)、深度烧伤患者40例(深Ⅱ度烧伤20例和Ⅲ度烧伤20例)、40例正常人的血清各分为预备组和检测组。预备组血清与金属亲和表面芯片结合后,用蛋白芯片仪读取数据,用CipherGen公司提供的BioMarker Wizard及BioMarker Pattern软件分析可得到用于区分浅度烧伤患者、深度烧伤患者及正常人血清的树状分类规则,并用双盲法测试检测组以证实其准确性。结果软件分析结果显示预备组中三类血清蛋白质在质荷比为5375Da,5717Da,4529Da,1225Da,1067Da,3883Da,66346Da 7处含量有显著差异,得到树状分类规则,其分类准确率为100%(120/120),灵敏度为100%(80/80),特异性为100%(40/40);对检测组进行双盲检测,结果显示准确度为100%(120/120),灵敏度和特异性分别为100%(80/80)、100%(40/40)。结论用SELDI-TOF-MS技术对正常人、浅度烧伤患者及深度烧伤患者进行比较蛋白质组学研究,发现在烧伤病理发展中有一组蛋白质有显著的差异。
Objective At present mechanism of adustum scar is not clear. This study aimed at establishing a serum protein fingerprinting technique coupled with a pattern - matching algorithm to distinguish adustum scar patients from adustum patients and healthy people. Methods Preliminary group and test group were both random selected serum samples, which each comprises 40 cases of adustum patient including 20 cases of degree Ⅰ adustum and 20 cases of superficial degree Ⅱ adustum, 40 cases of adustum scar patient including 20 cases of deep degree Ⅲ adustum and 20 cases of superficial degree Ⅱ adustum, 40cases of healthy people. The sera of the test group were combined with the surface of the IMAC3 proteinchip. Then reading data of SELDI - TOF - MS was analyzed by Biomarker Wizard software and Biomarker pattern software to get a classification rule of tree, which is standard configuration that can distinguish the sera of adustum scar patients from the sera of healthy people and adustum patients, and the standard was approved test validity by double - blindly in the test group. Results At the key M/Z values of 5375Da,5717Da,4529Da, 1225Da, 1067Da,3883Da,66346Da protein contents of the three classes in preliminary group are obviously different by the software analysis. And the classification rule of tree was discovered, and corresponding correct ratio was 100% (120/120), corresponding sensitivity was 100% (80/80) and corresponding specificity was 100% (40/40) ; After double - blind examining the test group with that rule, the corresponding correct ration was 100% ( 120/120), the corresponding sensitivity was 100% (80/80) and the corresponding specificity was 100% (40/40). Conclusion Via comparative proteomics research in the sera of adustum patients and adustum scar patients and healthy people, the group of protein was obviously different in adustum pathological process.
