摘要
目的:探讨克服肿瘤细胞多药耐药(MDR)的方法,提高化疗效果。方法:采用多药耐药反义基因(mdr-1-ASPS-ODN)逆转K562/ADM肿瘤细胞的MDR,而诱导肿瘤细胞凋亡。结果:mdr-1-ASPS-ODN诱导K562/ADM细胞产生大量DNA断片,流式细胞仪检测发现几乎全部mdr-1+K562/ADM细胞发生凋亡。结论:mdr-1-ASPS-ODN能有效、特异地抑制mdr-1基因表达,逆转肿瘤细胞的MDR,促进阿霉素诱导K562/ADM细胞凋亡,为其临床应用提供理论依据。
Objective:To explore an approach to the reversal of multidrug resistance(MDR) of tumor cells.Methods:Antisense oligodeoxynucleotide(mdr1AS PSODN) was used.Results:A large amount of DNA fragments was found in mdr1AS PSODN treated K562/ADM cells and almost all the mdr1+ K562/ADM cells underwent apoptosis.Conclusion: mdr1AS PSODN could inhibit specifically mdr1 gene expression,effectively reverse MDR of the tumor cells and promote ADM induced apoptosis of mdr1+ K562/ADM.
