应用荧光原位杂交方法检测鼻咽癌细胞系中EGFR基因拷贝数变化

摘要 目的检测鼻咽癌细胞系中表皮生长因子受体(EGFR)基因拷贝数扩增情况。方法以人鼻咽癌细胞系CNE-1、CNE-2、HONE-1、SUNE-1、HNE-1、HNE-2作为研究对象。应用探针LSI EGFR Spectrum Orange/CEP 7 Spectrum Green probe(Vysis,Abbott Laboratoriesm,IL)以荧光原位杂交(FISH)方法进行EGFR基因拷贝数的检测。结果经FISH法检测的6株鼻咽癌细胞株中,HNE-2存在基因扩增,其余细胞系为三倍体或多倍体。结论鼻咽癌细胞系中存在EGFR基因拷贝数的扩增,能否作为酪氨酸激酶抑制剂有效的分子标记,值得进一步研究。 Objective To detect epidermal growth factor receptor gene amplification in nasopharyngeal carcinoma cell lines.Methods Gene copy number per cell was investigated by fluorescence in situ hybridization(FISH) using the LSI EGFR SpectrumOrange/CEP 7 SpectrumGreen probe(Vysis,Abbott Laboratoriesm,IL) in nasopharyngeal carcinoma cells including CNE-1、CNE-2、HONE-1、 SUNE-1、HNE-1、HNE-2.Results By FISH,EGFR gene amplification was demonstrated only in HNE-2 cell;the other five cell lines were triploid or polyploid.Conclusion Our data indicate that there is EGFR gene amplification in nasopharyngeal carcinoma cells.Further study is warranted to find if EGFR gene amplification would be an effective molecular mark for tyrosine kinase inhibitors(TKIs).
