摘要
应用细胞融合技术将小鼠MHC基因引入人胃癌细胞,建立了1株杂交瘤细胞系。经染色体分析、流式细胞仪及ABC免疫组化染色证实,其具有杂交瘤细胞的特征,可同时表达人胃癌抗原和小鼠MHC分子。免疫学特性研究显示,该杂交瘤细胞能被小鼠肿瘤特异性杀伤细胞所识别和杀灭,有可能成为人胃癌疫苗研究模型中较理想的靶细胞。本项研究结果的意义是,在应用动物模型研究人类肿瘤抗原及疫苗时,进行种间细胞融合可能是克服靶细胞与效应细胞之间MHC不匹配的简单而有效的方法。
A human gastric cancer cell line with H2d gene was established by cell fusion between human gastric cancer cells and spleen cells of Balb/c mouse. In addition to MGAgs, the hybrid cells express MHC of Balb/c mouse, demonstrated by FACS and ABCaffinity cytochemistry analysis. Immunologic characteristic analysis showed that the hybrid could be recognized and killed by CTL (cytotoxic T lymphocytes) of Balb/c mouse. Thus a test model for the research upon human gastric cancer antigens and vaccines was established, in which the hybridoma cells act as target cells. These findings suggest an approach for overcoming the unmatched MHC between target cells and effective cells when we research human tumor antigens and vaccines in animal model system.
