摘要
目的研究高血糖素样肽-2(Glucagon-like peptide-2,GLP-2)对大鼠颅脑外伤后胆汁中分泌型IgA(sIgA)、肠粘膜T细胞及其亚群以及肠粘膜蛋白质和DNA含量的影响。方法制作大鼠弥漫性脑创伤模型,连续腹腔注射GLP-2,7d后收集大鼠胆汁测定sIgA、采集门静脉血测定血内毒素及制备小肠粘膜标本,检测小肠粘膜蛋白质、DNA含量和小肠上皮内淋巴细胞的T细胞及其亚群。结果颅脑外伤组大鼠小肠粘膜蛋白质和DNA含量以及胆汁中sIgA显著下降(P<0.01),小肠粘膜CD4+、CD4+/CD8+较对照组显著降低(P<0.05);而GLP-2组大鼠小肠粘膜蛋白质和DNA含量以及胆汁中sIgA明显增加(P<0.01),小肠粘膜CD4+、CD4+/CD8+较颅脑外伤组增加(P<0.05),门静脉血内毒素也显著减少(P<0.01)。结论GLP-2能够改善大鼠颅脑外伤后肠粘膜免疫功能,从而减轻内毒血症。
Objective To investigate the effect of GLP - 2 on the concentration of sIgA in bile, the changes of intestinal intraepithelial lymphocyte, content of protein and DNA in intestinal mucosa in rats following diffuse brain injury (DBI). Methods The animal model established by Marmarou was used to produced DBI. GLP -2 was injected twice daily for up to 7 days through intraperitoneal. We collected biles for measuring slgA, portal blood for exdotoxin andprepared intestinal mucosal specimens for measuring protein and DNA content and intestinal intraepithelial lymphocyte. Results The DNA and protein content in intestinal mucosa and the concentration of sIgA in bile of the DBI group was significantly reduced,There was a significant reduction in the number of CD^4+ T lymphocytes in rat intestines of the DBI group,and the ratio of CD^4+ to CD^8+ was decreased. The DNA and protein content in intestinal mucosa and the concentration of slgA in bile of the GLP - 2 group was significantly higher, the number of CD^4+ T lymphocytes in rat intestines of the DBI group and the ratio of CD^4+ to CD^8+ was increased in GLP - 2 group compared with DBI group. The portal endotoxin of GLP - 2 group was lower than DBI group. Conclusions GLP - 2 can improve the intestinal immunity of rat following DBI, then lead to relieved endotoxemia.
