二氢卟吩e4甘氨酸酰胺对小鼠S180移植肉瘤的光动力学效应

摘要 目的探讨630 nm及661 nm激光激发不同剂量的二氢卟吩e4甘氨酸酰胺(e4),对荷S180肉瘤小鼠的光动力学效应。方法昆明小鼠54只单侧腋下接种S180肉瘤,待肿瘤长至直径为6~8 mm时,随机分为空白对照组和PDT组。PDT组又分设e4 5 mg/kg和10 mg/kg两剂量组。小鼠尾静脉注射光敏剂后3 h,分别用630 nm或661 nm激光以150 mW/cm^2的功率密度垂直照射瘤体20 min,即能量密度180 J/cm^2,光斑直径1.5 cm。照光1周后取瘤组织称重并计算抑瘤率。结果630 nm和661 nm激光照射组均表现出光动力抑瘤效果。经630 nm激光照射,e4 5 mg/kg和10 mg/kg组的抑瘤率分别为35.5%和39.0%,两者差异无显著性意义(P>0.05)。而经661 nm激光照射,e4 5 mg/kg和10 mg/kg组的抑瘤率分别为50.49%及61.18%,两者差异有显著性意义(P<0.01)。而在同一e4浓度下,波长661 nm组较630 nm组的抑瘤率高(P<0.01)。结论二氢卟吩e4甘氨酸酰胺-PDT对小鼠S180肉瘤有明确的抑瘤作用,且661 nm激光的动力学抑瘤效应大于630 nm激光。 Objective Chlorin e4 glycine amide, a new type of photosensitizer, made from chlorophyll of silkworm excrement, has been developed by College of Pharmacy of the Second Military Medical University. The purpose of this study is to evaluate the photochemotherapeutic effects of this compound on sarcoma 180 transplanted in mice. Methods Subcutaneous inoculation of sarcoma 180 was made on double or single armpit of the both sexes Kunming mice. When the tumors reached 6-8 mm in diameter, the tumor transplanted mice were randomly divided into several groups according to weight and diameter of tumor. Tumors were irradiated vertically with 630 nm ( semiconductor laser) for 20 min at a power density of 150 mW/cm^2 [ energy density(ED) : 180 J/cm^2 ] at 3 hours after intravenous injection in a dose of 5 mg/kg or 10 mg/kg. The tumor weight was deter-mined and the inhibiting rate of tumor was calculated 1 week after laser irradiation. Results Chlorin e4 glycine amide could obviously inhibit the sarcoma growth, the inhibiting rate of tumor in 5 mg/kg and 10 mg/kg groups after 630 nm irradiation was 35.5% and 39. 0% , but after 661 nm irradiation it was 50. 49% and 61.18% ,respectively. Conclusions Chlorin e4 glycine amide has obvious anti-sarcoma effect on S180 sarcoma-bearing mice.
