中药对去势大鼠神经内分泌免疫网络的影响 被引量:13

Effect of Traditional Chinese Medcine on Receptor-neuroendocrine-immune Network in Ovariectomized Rats

摘要 [目的]从神经-内分泌-免疫网络(RNEI-N)角度探讨补肾活血中药复方益妇宁软胶囊(YFN)改善更年期综合征(CS)的作用机制。[方法]采用大鼠去势复制CS动物模型,给药4周后,测定大鼠血清E2、FSH、LH、Te、P、IL-2及下丘脑、垂体和血浆β-EP水平;测定下丘脑5-HT、5-5-HIAA、NE及DA含量;检测子宫、脾脏和主动脉雌激素受体(ER)水平。[结果]YFN能升高去势大鼠血清E2、IL-2水平,提高子宫、脾脏和主动脉ER水平,升高下丘脑、垂体和血浆β-EP水平,降低下丘脑5-HT和5-HIAA水平,升高NE和DA的含量,降低5-HT/NE、5-HT/DA的比值。[结论]YFN通过调节生殖内分泌功能,改善植物神经功能紊乱,提高机体ER水平和免疫功能,能从多层次、多途径、多环节作用于更年期衰退的神经内分泌免疫网络,从而达到稳定机体内环境,缓解CS症状的作用。 [Objective]To explore the therapeutic mechanisms of traditional Chinese medicine formula Yifuning soft capsules(YFN) which can nourish the kidney and activate blood on treating climacteric syndrome(CS)from neuroendocrine- immune network(NEIN). [Method]We established the models of climacteric syndrome with ovariectomized(OVX)rats. After giving drugs 4 weeks, we detected serum sex hormone (E2, P, Te, FSH, LH), interleukin-2 ( IL-2 ) and hypothalamic, pituitary and plasma β-endorphin (β-EP) levels. The hypothalamic monoamine transmitters (NE,DA, 5-HT, 5-HIAA) contents were detected. The estrogen receptor (ER) levels of uterus,spleen and artery were detected too. [Result]YFN can. improve levels of serum E2,IL-2 and ER of uterus, spleen and artery in OVX rats. YFN can increase reduced hypothalamic,pituitary and plasma β-EP levels in OVX rats. YFN can reduce contents of hypothalamic 5-HT and 5-HIAA,increase the levels of NE and DA in OVX rats and reduce the ratio of 5-HT to NE and 5-HT to DA in OVX rats. [Conclusion]YFN may produce effects on declined NEI-N in menopause from various levels, ways and links by means of regulating the function of plant nerve and reproduction endocrine and improving levels of ER and body immunity. So they can stabilize inner condition of body and relieve the syndrome of CS.
