摘要
目的从分子水平探讨大鼠局灶性脑缺血再灌注后的胶质细胞源性神经营养因子mRNA(GDNF mRNA)的表达规律性及人参皂甙Rb1对其的影响。方法健康成年Wistar大鼠70只,随机分成4组:正常组、假手术组、单纯脑缺血再灌注组和Rb1治疗组。用线栓法阻塞大鼠大脑中动脉(MCA)2 h再灌注3 h^5 d制备脑缺血模型,在脑缺血2 h再灌注后3 h^5 d的各时间点取材;逆转录-聚合酶链反应(RT-PCR)测定GDNF mRNA水平变化。结果脑缺血再灌注损伤后GDNF mRNA表达在3 h^2 d分别有2个高峰。给予人参皂甙Rb1后,GDNF mRNA表达在2 d达高峰。根据2组间GDNF mRNA量的比较,给予人参皂甙Rb1后,在同一时间点,GDNF mRNA的量明显增多。对GDNF mRNA的量统计分析显示,Rb1给药组各时间点GDNF mR-NA的量远远高于单纯脑缺血再灌注组(P<0.01),提示人参皂甙Rb1诱导GDNF mRNA表达。结论脑缺血再灌注后GDNF mRNA的表达与缺血性损伤有一定的联系,人参皂甙Rb1对中枢神经系统有保护作用。
Objective It is to discuss the expression regularity of GDNF mRNA in focal cerebral ischemia rats after reperfusion on molecular level and the influence of ginsenoside Rbl on it. Methods Seventy healthy Wistar rats were randomly divided into normal group, sham operated group, simple cerebral ischemia reperfusion group and Rbl treatment group. The cerebral ischemia model was established through obstructing middle cerebral artery of rats for 2 h with line-cork method and reperfusing 3 h to 5 d. The materials were drawn from every time point of cerebral ischemia 2 h after reperfusing 3 h to 5 d. The change of GDNF mRNA level was determined with RT- PCR. Results The expression of GDNF mRNA had two peaks at 3 h to 2 d after cerebral ischemia reperfusion injury. After ginsenoside Rbl was given, the expression of GDNF mRNA peaked at 2 d. The content of GDNF mRNA at the same time point was obviously increased after ginsenoside Rbl was given. The content of GDNF mRNA in Rbl treatment group at every time point was more higher than that in simple cerebral ischemia reperfusion group ( P〈 0.01 ), which prompted that ginsenoside Rbl induced GDNF mRNA expression. Conclusion The expression of GDNF mRNA after cerebral ischemia reperfusion is related to ischemic injury. Ginsenoside Rbl has protective action on central nervous system.
