木犀草素-7-O-β-D-葡萄糖苷对缺血缺氧培养乳鼠心肌细胞的保护作用 被引量:17

摘要 目的观察木犀草素-7-O-β-D-葡萄糖苷对缺血缺氧培养乳鼠心肌细胞的保护作用及其可能机制。方法采用原代培养的乳鼠心肌细胞造成缺血缺氧损伤模型,观察木犀草素-7-O-β-D-葡萄糖苷对损伤后心肌细胞存活率、超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量的影响,推断细胞内活性氧的生成量;观察培养液中乳酸脱氢酶(LDH)活性,以判断细胞损伤情况;并进行hochest33258染色,判断细胞凋亡程度。结果木犀草素-7-O-β-D-葡萄糖苷(100,50μg/mL)组可显著升高缺血缺氧损伤时细胞存活率、提高细胞SOD活性,降低MDA含量、降低LDH的漏出量,细胞凋亡率降低。结论木犀草素-7-O-β-D-葡萄糖苷对缺血缺氧培养的心肌细胞具有保护作用,其机制可能与清除氧自由基,稳定细胞膜,抑制细胞凋亡有关。 Objective To study the protective effects of luteolin -7-O-β-D- glucopyranoside on myocardial cells injury induced by hypoxia in neonatal rats and to explore its mechanism. Methods The model of isehemie injury of primary cultured myocardial cells of neonatal rats was established to investigate the effects of luteolin-7-O-β-D- glucopyranoside on the in- jury. Then lactate dehydrogenase (LDH)and superoxide dismutase (SOD)malondialdehyde (MDA) were determined in each group. Cell apoptosis rate was identified by hechest33258 staining. Results Luteolin-7-O-β-D-glucopyranoside ( 100, 50 μg/mL)could increase the survival rate of the cells with hypoxic - ischemic injury and SOD activity, decrease MDA content, reduce the leaking of LDH and alleviate the cell apoptosis rate. Conclusions Luteolin-7-O-β-D-glucopyranoside has protective effect on ischemic injury, which may be related to the stabilizing of cell membrane, the clearance of oxygen free radicals and the inhibition of cell apoptosis.
