摘要
目的研究多药耐药相关蛋白1(multidrug resistance-associated protein1,MRP1)在大肠癌中的表达及其与临床病理指标的关系,为大肠癌的有效防治提供科学依据。方法应用免疫组织化学SP法对53例大肠癌患者的原发灶癌组织、距原发灶10cm经病理证实的正常组织进行MRP1蛋白免疫组织化学检测,并将检测结果与患者的年龄、性别、肿瘤大小、淋巴结转移、肿瘤浸润深度、分化程度、生长部位、Dukes分期情况进行综合分析。结果MRP1蛋白主要是在细胞膜和(或)细胞浆内表达,MRP1蛋白在大肠癌原发灶组、正常组织组的阳性表达率分别为49.1%(26/53)和15.1%(8/53),两者有显著性差异(P<0.01)。MRP1的表达与大肠癌组织的分化程度、浸润深度、淋巴结转移等病理特征均无关。结论MRP1可能与大肠癌的发生有关,并与原发性多药耐药性有关,MRP1可能是独立于病理学特征之外的分子参数。
Objective To investigate the expression of multidrug resistance-associated protein I(MRP1) in colorectal carcinoma and the relationship with clinicopathological factors to provide scientific basis for effective prevention and cure of colorectal carcinoma. Methods Fresh samples were obtained from 53 patients who underwent colorectal carcinoma operation. For each patient, 2 samples were obtained, which were respectively from the loci of carcinoma and colorectal tissue 10cm away from the loci of carcinoma that proved to be normal colorectal tissue by pathology. The expression of MRP1 were studied by means of immunohistochemical assay (SP method). The level of the expression was correlated with clinicopathological factors. Results MRP1 was mainly expressed on carcinoma cell membrane and (or) cytoplasm,and in the positive expression cell,the membrane and (or) cytoplasm was buffy. The positive expression rate of MRP1 in the loci of carcinoma group and normal colorectal tissue group were 49.1% (26/53) and 15.1% (8/53) respectively. The positive expression rate of MRP1 in the loci of carcinoma tissue was higher than that of normal colorectal tissue. There was a significant difference betweenthe two groups ( P 〈0.01). The expression of MRP1 has no correlation with the degree of differentiation, the depth of invasion, the metastasis of lymph node and all the otherer clinicopathological factors ( P〉0.05). Conclusion The overexpression of MRP1 protein may involve in colorectal carcinogenesis and the primary multidrug resistance in colorectal carcinoma. MRP1 may be an independent molecular prognostic index without correlation between their expressions and pathological characteristics.
