摘要
目的探讨肠黏膜异常隐窝灶(ACF)与正常肠黏膜、腺瘤及腺癌中APC、β-catenin、K-ras蛋白表达的差异。方法采用免疫组化Envision法检测ACF、正常肠黏膜、腺瘤及腺癌组织中APC、β-catenin、K-ras蛋白表达。结果异常增生型ACF与正常肠黏膜相比较,APC、K-ras蛋白表达差异具有统计学意义(P<0.05),β-catenin表达差异具有统计学意义(P<0.01);与腺瘤相比较,β-catenin表达差异具有统计学意义(P<0.05),APC、K-ras表达差异无统计学意义(P>0.05);与腺癌比较,APC、β-catenin、K-ras蛋白表达差异具有统计学意义(P<0.01)。增生型ACF与正常肠黏膜比较,APC、K-ras蛋白表达差异具有统计学意义(P<0.05),β-catenin表达差异无统计学意义(P>0.05);与腺瘤比较,β-cate-nin表达差异具有统计学意义(P<0.05),K-ras表达差异具有统计学意义(P<0.01);与腺癌比较APC、β-catenin、K-ras表达差异具有统计学意义(P<0.01)。异常增生型ACF、腺癌中K-ras蛋白阳性表达与β-catenin蛋白异位表达呈正相关(P<0.05),APC蛋白阳性表达与K-ras蛋白阳性表达呈负相关(P<0.05)。结论ACF作为最早期的癌前病变具有异常的蛋白表达,异常增生型ACF尤为明显。APC、β-catenin、K-ras蛋白表达异常相互作用促进了大肠肿瘤的发生。
Objective To study the differences of APC, β-catenin,K-ras expression in aberrant crypt foci (ACF), normal intestinal crypt foci,adenoma and adenocarcinoma. Methods Immunohistocbemistry by Envision was used to detected APC, β-catenin,Kras expression in ACF, the normal intestinal, adenoma and adenocarcinoma. Results The differences of APC, K-ras, β- catenin were significant between abnormal hyperplastic ACF and normal gut (P 〈 0.05 or P 〈 0.01 ) ;Compared with adenoma, β- catenin expression was significant difference( P 〈 0.05 ), APC, K-ras had no difference (P 〉 0.05 ) ;Compared with adenocarcinoma, expressions of APC, β-catenin, K-ras were significant difference (P 〈 0.01 ). Differences of APC, K-ras expression were significant between hyperplastic ACF and normal gut( P 〈 0.05 ), β-catenin expression has no difference between them( P 〉 0.05 ) ; Compared with the adenomas, β-catenin and K-ras expression had significant differences(P 〈 0.05 or P 〈 0.01 ) ;Compared with adenocarcinoma, ACF had different expressions of APC, β-catenin, K-ras (P 〈 0.01 ). Positive expression of K-ras in abnormal hyperplastic ACF was positively correlated with ectopic expression of β-catenin in adenocarcinoma. Positive expression of APC was negative correlated with expression of K-ras ( P 〈 0.05 ). Conclusions As the earliest precancerous lesions, ACF has abnormal protein expression, dysplasia of ACF is particularly evident. Abnormal protein expression of APC, β-catenin, K-ras promote the occurrence of cancer of colon.
