目的:研究胃癌细胞在右旋甲硫氨酸(D-Met)环境中氨基酸的代谢。方法:将6株胃癌细胞分别接种于D-Met、Met-和左旋甲硫氨酸(L-Met)培养液中,培养5 d后,用高效液相色谱法检测培养液中20种氨基酸的浓度。结果:Met-组6株胃癌细胞的Met含量显著低于L-Met组和D-Met组(P<0.05);D-Met组和Met-组SUN-1、KATO-Ⅲ和MKN45细胞的同型半胱氨酸(Hcy)含量显著高于L-Met组(P<0.05);除SUN-1和MKN45外,D-Met组和Met-组各株细胞Gln含量显著高于L-Met组(P<0.05);D-Met组和Met-组KATO-Ⅲ、NCI-N87和AGS的丙氨酸(Ala)含量显著低于L-Met组(P<0.05);D-Met组和Met-组SUN-16、KATO-Ⅲ和AGS的半胱氨酸(Cys)含量显著低于L-Met组(P<0.05)。结论:D-Met环境可影响胃癌细胞代谢,其效果与Met-环境相似。
Objective : To investigate the amino acids metabolism of gastric cancer cells in D-methionine (D-Met) medium. Methods : Six gastric cancer cell lines were respectively cultured with L-methionine ( L-Met), D-Met, or without methionine (Meta^-) medium. Amino acids profile of every culture medium was measured by HPLC after 5 days. Results : Methionine concentration of six cell lines in Met-medium was significantly lower than that in L-Met or D-Met medium (P〈0.05). Homocysteine concentration of SUN-1, KATO-Ⅲ, and MKN45 cell lines in D-Met or Met medium was significantly higher than that in L-Met medium(P 〈0.05). Glutamine concentration of all cell lines but SUN-1 and MKN45 in D-Met or Met-group was significantly higher than that in L-Met medium(P〈0.05). Alanine concentration of KATO-Ⅲ ,NCI-N87 and AGS cell lines in D-Met or Met^- group was significantly lower than that in L-Met medium(P〈0.05). Cysteine concentration of SUN-16, KATO-Ⅲ, and AGS cell lines in D-Met or Met medium was significantly lower than that in L-Met medium(P〈0.05). Conclusion : D-methio-nine environment could affect amino acids metabolism of gastric cancer cells, and its effect was similar to Met-environment.
