摘要
目的:探讨HIF-1α、VEGF在胃癌组织中的表达及相关性。方法:应用免疫组织化学方法检测84例胃癌组织及癌旁正常组织中HIF-1α、VEGF的表达,并分析它们与临床病理因素的关系及相互间的关系。结果:84例胃癌组织中及癌旁内对照正常胃组织HIF-1α阳性表达分别为60.7%,16.7%(P<0.05)。有、无淋巴结转移的癌组织中HIF-1α的表达率分别为79.1%、41.5%(P<0.05),侵润深度分别为粘膜或粘膜下、肌层或浆膜下、穿透浆膜层的癌组织中HIF-1α的表达率分别为29.2%、62.2%、91.3%(P<0.05)。未或低分化、中或高分化的癌组织中HIF-1α的表达率分别为76%、54.2%(P>0.05)。肿瘤大小≤5cm、>5cm的癌组织中HIF-1α的阳性表达率分别为64.7%、58%(P>0.05)。男性为63%,女性为57.9%(P>0.05),<60岁为52.3%,≥60岁为70%(P>0.05)。VEGF阳性表达为73.8%(62/84)。HIF-1α与VEGF的表达显出相关性,P<0.05。结论:HIF-1α与胃癌的侵润、淋巴结转移有关,并与VEGF的表达具有明显相关性,HIF-1α的作用部分通过VEGF的表达实现。因此可以将HIF-1α作为胃癌患者判断生物学行为及预后的指标之一,也为临床提供靶向治疗的方向。
Objective:To evaluate the expressions of HIF-1αand VEGF in gastric cancers,and explore the correlation between HIF-1αand VEGF. Methods:The expressions of HIF-1αand VEGF in 84 specimens of gastric cancer and paracancerous normal tissues were detected by SP immunohistochemistry and the correlations of HIF-1α and VEGF to clinicopathologic features and association between HIF-1α and VEGF were analyzed with Sas 8.0 software. Results:Of the 84 specimens,the positive rate of HIF-1α expression was 60.7%(51/84),and it was much higher than that in paracancerous normal tissues (16.7%,14/84,P〈0.05). HIF-1α expression in tissues with lymph node metastasis(79.1%) was much higher than that in tissues without no lymph node metastasis 41.5%(P〈0.05). The expression rates of HIF-1α in cancer tissues with the invasion depth as the mucous membrane or under,muscle layer or under serosa,and penetrating the serosa were 29.2%,62.2% and 91.3% respectively(P〈0.05). Expression rates of HIF-1α in cancer tissues with no or low differentiation,and medium or high differentiation were 76% and 54.2% respectively(P〉0.05). Expression rates in cancer tissues with tumor size≤5cm and 〉5cm were 64.7% and 58% respectively(P〈0.05),The rate for male was 63% while that for female was 59.7%(P〉0.05),The rate for patients aged 〈60 was 52.3%,while that for those aged≥60 was 70%(P〉0.05)o The positive rate of VEGF expression was 73.8%(62/84). The expression of VEGF was significantly associated with that of HIF-1α (P〈O.05). Conclusion:The expression of HIF-1α is correlated to lymph node metastasis and the depth of invasion,The function of HIF-1α realizes partially through the expression of VEGF. Therefore,HIF-1α could serve as a parameter for evaluating malignancy and prediction of prognosis of gastric cancer and HIF-1α may be a direction of clinical therapy.
