农杆菌介导的几丁质酶基因转化花生的研究 被引量:4

摘要 以花生品种花育20成熟胚萌发5~7d的幼叶为外植体,利用农杆菌介导法进行几丁质酶基因(CH5B)的遗传转化,并对转化条件进行了研究。结果表明,外植体经预培养2d,于OD600=0.6~0.8的农杆菌(GV3101)菌液中浸泡15min,再共培养3d,最适宜遗传转化。对抗性植株进行PCR检测,扩增得到了目的条带,表明几丁质酶基因成功转入了花生。 CH5B gene was transferred into peanut cultivar Huayu No. 20 mediated by Agrobacterium tumefaciens , and transformation conditions were studied in this paper. The result showed that pre - culture for 2 days, Agrobacterium suspension (OD600 =0.6-0.8) for 15 min and co-culture for 3 days was suitable to obtain resistant shoots. The target band of resistant plants was amplified by PCR analysis. These results confirmed that CH5B gene was transferred into peanut successfully.
