摘要
目的评价长春新碱传递体(vincristine loaded transfersom es,VCR-T)对荷Burk itt’s淋巴瘤裸鼠的抑瘤效果。方法建立荷Burk itt’s淋巴瘤的裸鼠模型,将VCR-T分为高(0.8 mg.kg-1)、中(0.2 mg.kg-1)、低(0.05 mg.kg-1)剂量以及单次给药和3次给药组,长春新碱溶液(vincristine solution,VCR-Sol)以0.8 mg.kg-1的剂量分3次静脉注射,比较各组荷瘤裸鼠的相对肿瘤增殖率、肿瘤生长曲线、抑瘤率,并进行瘤体组织学检查考察VCR-T的抑瘤效果。结果各治疗组对肿瘤的生长均有一定的抑制作用,且随着给药剂量及给药次数的增加,对肿瘤的抑制作用也相应的增加;VCR-T以0.8 mg.kg-1剂量单次给药则可达到与VCR-Sol相同的效果,而0.2 mg.kg-1剂量3次给药的疗效则优于VCR-Sol;所有VCR-T组裸鼠100%存活,VCR-Sol组裸鼠存活率仅75%。结论VCR-T透皮给药具有比VCR-Sol静注给药更好的抑瘤效果。
OBJECTIVE To investigate the tumor inhibition effect of vincristine loaded transfersomes (VCR-T) on Burkitt' s lymphoma in nude mice. METHODS The Burkitt' s lymphoma model of Blab/C nude mice was made to evaluate the therapeutic effect of VCR-T by external administration. The mice were divided into 6 groups treated with the single- and multiple-dose at the high, middle and low levels(0. 8 , 0.2, 0.05 mg·kg^-1 ). The reference preparation was vincristine solution(VCR-Sol) at doses of 0. 8 mg·kg^-1. The relative tumor proliferation rate, growth curves, inhibition rate and histological examination were obtained. RESULTS All group showed an inhibition effect on the growth of tumor, and there was dose-effect relationship in the treatment of VCR-T. VCR-T reached the same effect as VCR-Sol at dosage of 0. 8 mg·kg^-1 with single administration, and got superior results after three administration of 0. 2 mg·kg^-1. Besides, the survival rate of mice treated by VCR-T was 100%, and 75% by VCR-sol. CONCLUSION VCR transfersomes get better inhibition efficiency than VCR-sol.
