重组人睫状神经营养因子对糖尿病大鼠胃肠功能的影响 被引量:7

Effects of Recombinant Human Ciliary Neurotrophic Factor on Intestine Function in Diabetes Mellitus Rat Model

摘要 目的评价重组人睫状神经营养因子(rhCNTF)对糖尿病大鼠胃肠道功能的作用。方法采用链脲佐菌素(strepto- zocin,STZ)诱发的糖尿病大鼠胃肠神经功能紊乱模型,肌注rhCNTF 0.1,0.3,0.9mg·kg^(-1),测定了rhCNTF对血糖、胃肠慢波电位、小肠壁内神经蛋白含量,观察了rhCNTF对胃肠推进功能的影响以及胃肠对乙酰胆碱(Ach)、阿托品敏感性的影响。结果肌内注射rhCNTF对血糖无明显影响,也未见rhCNTF改善十二指肠对Ach、阿托品的敏感性。肌注RhC- NTF可改善糖尿病大鼠胃肠慢波电位、提高了胃肠推进功能、增加了胃肠壁内神经蛋白含量。结论糖尿病导致的胃肠神经功能紊乱可以通过肌注rhCNTF而改善,其重要机制是rhCNTF对抗糖尿病引起的胃肠神经病变。 OBJECTIVE To investigate the activity of the recombinant human ciliary neurotrophie factor (rhCNTF) on gastrointestinal neurosis in diabetic rats. METHODS Diabetic rat models were built by STZ, and rats were administered with rhCNTF for 85 d after the administration of streptozocin(STZ). The dose of RhCNTF were 0. 1 ,0. 3,0. 9 mg · kg-1. Effects of rhCNTF on blood glucose, slow wave in stomach intestine, S100 in intestine, peristalsis, sensitivity to Ach and artropine in intestine were determined. RESULTS The effect of rhCNTF on blood glucose was not obtained compared with the vehicle group, and so did the contract character of jejunum treated with Ach and Atropine. The frequency of slow wave of jejunum was improved. The drive functions of the intestine were augmented, and so did the contents of SI00 in intestine. CONCLUSION The function of intestine in diabetic rat can be improved by rhCNTF administering, and the important mechanism is the nerve protective effects of rhCNTF.
