摘要
目的研究裙带菜茎多糖体外抗单纯疱疹病毒1型(HSV-1)活性。方法采用空斑减数实验测定裙带菜茎粗多糖及纯化后多糖抗HSV-1活性,计算它们的空斑抑制率和IC50,并从药物对病毒的直接灭活作用、对病毒吸附的影响及对病毒释放的影响3个方面初探裙带菜茎多糖抗单纯疱疹病毒活性机理。结果裙带菜茎多糖能明显抑制HSV-1的致病变作用,IC50依次为8.04,13.54μg/m l。药物对病毒没有直接灭活作用,对病毒的释放也没有影响,主要影响了病毒的吸附。结论裙带菜茎多糖有显著的抗HSV-1活性,且粗多糖比纯化后多糖活性要好,并且可以初步推测裙带菜茎多糖抗HSV-1活性是作用在病毒和受体结合、侵入Vero细胞阶段。
Objective To investigate the activities of anti - Herpes simplex 1 virus of polysaccharide extracted from the caudex of Undariapinnatifida. Methods HSV - 1 was inoculated on Vero cells, polysaccharide extracted from the caudex of Undaria pinnatifida was added to the medium in various concentrations. After 72 hours incubation, plaques were counted and the 50% inhibiting concentration (IC50) was calculated in Plaque reduction assay . Initial studies on antivirus mechanism in three aspects: virus inactivation, the drug influence on virus absorption and release were performed. Results Polysaecharide had conspicuous anti - Herpes simplex 1 virus activities, their IC50 were 8.04 p.g/ml, 13.54 μg/ml respectively. The drug had no influence on inactivating virus and release from the cell, however, it had influence on virus absorption. Conclusion Polysaccharides extracted from the candex of Undaria pinnatifidahas has conspicuous anti - Herpes simplex 1 virus activity, and the activity happens in the binding, absorpting stage of HSV - 1 release with Vero cell.
