摘要
目的研究玉郎伞多糖(YLS)对肝星状细胞(HSC)氧应激脂质过氧化作用的影响及机制。方法用H2O2诱导肝星状细胞脂质过氧化,测定细胞培养上清液中MDA,SOD水平,MTT法测定肝星状细胞的增殖,LDH法检测YLS对肝星状细胞的毒性作用,ELISA法测定I型胶原含量。结果YLS(25-200μg/ml)可明显降低培养上清液中MDA含量和I型胶原水平,抑制肝星状细胞的增殖,提高SOD活力,并呈剂量依赖性。结论YLS对肝星状细胞的增殖及I型胶原的合成具有抑制作用,该作用可能与其抗氧化作用有关。
Objective To study the effects and its mechanism of Yulangsan polysaccharide(YLS) on the proliferation and collagen Ⅰ production of hepatic stellate cell(HSC) induced by oxidative stress. Methods The lipid peroxidation of HSC was induced by H2 O2. The levels of malondialdehyde (MDA) and SOD in cultural supernatant were determined by general methods. Cytotoxicity was measured by LDH method. The proliferation of hepatic stellate cells was measured by MTT method. The content of collagen Ⅰ was measured by ELISA method. Results YLS (25 - 200μg/ml)concentration -dependently decreased the levels of MDA and collagen Ⅰ and the number of proliferating HSC ,and increased the activity of SOD . Conclusion YLS can inhibit the proliferation and collagen Ⅰ production of hepatic stellate cell induced by oxidative stress. This might be associated with the anti - oxidative activity of YLS.
