摘要
目的对酶法提取山茱萸总皂苷的酶解条件进行了初步研究,考察了酶解时间、酶用量、酶解温度和酶解初始pH值对总皂苷产量的影响。方法采用紫外可见分光光度法测定总皂苷含量,检测波长为554 nm。结果确定了酶解的最优条件为:酶解时间为150 min,酶量为5 U/g,酶解温度为45℃,酶解初始pH=5.4。结论酶法能显著提高山茱萸总皂苷的提取率。
Objective A new technology of extracting total saponins from Fructus Corni with cellulose enzyme was studied in this experiment. The influence of enzymolysis time, enzyme amount, enzymolysis temperature, and pH of the enzyme solution was studied. Methods The content of total saponins in Fructus Corni determined by UV - speetrophoto - metry with a detection wave- length at 554 nm. Results The experiment showed an optimum process: enzymolysis time was 150 min, enzyme amount was 5U/ g, enzymolysis temperature was 45℃ and pH of enzyme solution was 5.4. Conclusion The extraction percentage of total saponins from Fruetus Corni can be improved by cellulose enzyme treatment.
