大孔吸附树脂分离纯化黄连、关黄柏中季铵总碱的工艺研究 被引量:11

摘要 目的利用AB-8大孔吸附树脂纯化黄连、关黄柏中季铵总碱,确定树脂纯化季铵总碱的工艺参数。方法以季铵总碱和盐酸小檗碱吸附量为指标,并通过正交实验考察确定了该树脂分离纯化季铵总碱的工艺条件。结果AB-8型树脂对季铵总碱有良好吸附分离性能,其吸附分离季铵总碱的工艺条件为:上样浓度为50 mg/m l(生药量),上样液pH值为8,吸附流速为3BV/h,上样体积为5BV,洗脱剂为5倍量树脂柱体积50%乙醇。结论AB-8型大孔吸附树脂在所确定的工艺条件下,纯化季铵总碱效果良好,季铵总碱的纯度可达90%。 Objective To set up a method for purification of total quaternary ammonium hydroxide from Coptis chinensis Franch. and Phellohendron amurense Rupr, by AB - 8 macroporous resin . Methods The technical process for purification of total quaternary ammonium based on AB -8 macroporous resin was selected by dynamic absorption ratio through orthogonal tests. Results The AB - 8 macroporous resin owned optimum absorption and elution ability. The optimum absorption conditions were that the sample concentration was 50mg · ml^-1, the PH of sample was at 8, the current velocity was 3BV· h^-1, the eluant was 50 % ethanol(SBV). Conclusion The AB - 8 macroporous resin can be used to purify total quatemary ammonium hydroxide successfully. The purity of total quatemary ammonium hydroxide is about 90 %,
