摘要
目的研究复方鳖甲软肝方对特发性肺间质纤维化防治的MMPs表达,探讨其机理和疗效。方法选SD雄性大鼠180只。随机分为假手术组、模型组、阳性药物对照组及复方鳖甲软肝方高、中、低剂量组,共6组,每组30只。各组分别于7,14,28d随机抽取10只大鼠,麻醉后取肺组织,MMP-2采用原位杂交及RT-PCR的方法测定。MMP-1和MMP-9采用免疫组化的方法。结果复方鳖甲软肝方组在早期抑制MMP-9在细胞内表达上调,在中晚期抑制MMP-1在细胞内表达上调,说明两者均有抑制纤维合成的作用。MMP-2的原位杂交和RT-PCR结果一致,在模型组过度表达,在药物组表达被抑制,表明药物对IPF的形成有抑制作用,以中、小剂量组为好。结论复方鳖甲软肝方在早期抑制MMP-9在细胞内表达上调,在中晚期抑制MMP-1在细胞内表达上调,达到抗纤维化作用;复方鳖甲软肝方通过抑制MMP-2表达发挥抗纤维化的作用。
Objective To study the effect of compound prescription witch turtle shell (CPTS) on the expression of MMPS in rats with IPF (Idiopathic pulmonary interstitial fibrosis) caused by Bleomycin AS. Methods 180 male SD rats were randomly divided into six groups:sham operation group, model group, positive medicine groups, high -dosage group, medium- dosage group and low - medium group. After they were given the medicine, ten rats in each group each time were killed respectively on the 7th day, 14th day and 28th day. MMP - 2 was detected by in situ hybridization and RT - PCR, MMP - 9 by immunohistochemical method. Results CPTS could inhibit expression of MMP - 9 in early phase and MMP - 1 in middle and late phases. Result of hybridization in situ of MMP - 2 was similar to that of RT - PCR. The effects of medium dose groups and low dose were better. Conclusion - CPTS can inhibit the expression of MMP - 9 in early phase and MMP - 1 in middle and late phases. CPTS has a protective effect on IPF by inhibiting the expression of MMP - 9.
