反相高效液相色谱法测定裸鼠肿瘤组织中苯并三唑衍生物(BZ)代谢产物的含量 被引量:1

摘要 建立了反相高效液相色谱(RP-HPLC)法测定裸鼠肿瘤组织中苯并三唑衍生物(BZ)代谢产物BZ-M1和BZ-M2的含量。以SNZ为内标,样品采用液-液萃取法处理。色谱柱为Dikma Diamonsil^(TM)C_(18),流动相为乙腈:1%冰醋酸=12:88,流速为1.0mL/min,检测波长为254nm。该法测定肿瘤组织中BZ-M1、BZ-M2的线性范围均为0.1~10.0μg/mL,BZ-M1萃取回收率为82.9%±3.3%。BZ-M2萃取回收率为86.3%±4.0%,其三批样品精密度RSD均小于10%。结果表明,该方法灵敏度高、准确度高、重复性好,可用于肿瘤组织中BZ的代谢产物BZ-M1和BZ-M2含量测定。 Objective : A HPLC method was developed for the determination of metabolites BZ - M1 and BZ - M2 in naked rat tissue. Methods : SNZ was used as internal standard. The sample was extracted by liquid - liquid extractive method, The analytical column was Dikma Diamonsil^TM C18, The mobile phase consisted of acetonitrile - 1% Acetic Acid ( 12 : 88 ). The detection wavelength was 254nm and the flow rate was 1.0mL/min. Results: Linear scopes of the metabolites BZ- M1 and BZ- M2 were in range from 0.1 -10.0μg/mL. The average extraction of recovery of BZ - M1 was 82.9% ± 3.3% , and the averager extroction of recovery of BZ - M2 was 86.3% ± 4, 0%. The RSD of three batch samples was less than 10%. Conclution: The method is high sensitivity, result is accurate and good reproducible. The analytic condition could be used to determine metabolites in naked rat tissue of BZ.
